If you're looking for T1 speeds and above, then you will need line of sight between your buildings. That being said, achieving line of sight is easier than you might think. Oftentimes it's a matter of going another ten or fifteen feet off a rooftop or perhaps trimming some trees back. Otherwise, we'd need to locate a structure (e.g., rooftop, water tank, tower, etc.) that can see both endpoints that you need to connect. That would be the "repeater" or "relay" site.
If you get up on your rooftops and notice several repeater possibilities, then consider which of those might be friendly to your cause. That might be the bank that processes your company's payroll or a university you do research with. In those cases you should expect speedier approval and a lower monthly roof space fee - if not a waiving of a fee altogether. We often find free or cut-rate roof space, particularly for educational and healthcare concerns.
Rooftops are always preferable to towers from an installation and servicing standpoint, so don't automatically canvas for towers if other options are present. For more information on determining line of sight, read: "How is line of sight determined".
